Aging is not only wear and tear — it is amnesia. The genome survives intact, but the chemical annotations that tell each cell which genes to run fade, smear and drift. Epigenetic restoration is the field asking the most audacious question in biology: if the pattern was written once, can it be written back? The "-100" is the aspiration — a century of healthy lifespan.

What epigenetic aging actually is

Every cell in your body carries the same genome. A neuron and a skin cell differ not in the book they own, but in which pages are bookmarked. The bookmarks are epigenetic marks — chiefly methyl groups stamped onto cytosines at CpG sites — and they are laid down, read and erased by dedicated molecular machinery. Aging, in the epigenetic view, is the slow corruption of that annotation layer.

The clock that started it all

In 2013, Steve Horvath at UCLA showed that methylation levels at just 353 CpG sites predict the biological age of virtually any human tissue — the "Horvath clock". For the first time, aging had a ruler. Every claim of rejuvenation since has had to answer to it.

Information loss, not just damage

David Sinclair's lab at Harvard Medical School argues (Cell, 2023) that disrupting the epigenome — without touching the DNA sequence — is itself sufficient to drive aging in mice. Break the filing system, and a young animal turns old. Restore it, and the old tissue remembers.

The reset button exists

In 2006 Shinya Yamanaka at Kyoto University converted adult cells back to stem cells with four factors — Oct4, Sox2, Klf4, c-Myc. Full reprogramming erases identity. The discovery that changed longevity science: partial reprogramming — a short pulse, then stop — resets age while the cell remains itself.

Restoration in living animals

Ocampo and colleagues in Juan Carlos Belmonte's lab at the Salk Institute (Cell, 2016) pulsed OSKM cyclically in mice and ameliorated age-associated hallmarks. Lu and colleagues in the Sinclair lab (Nature, 2020) restored vision in old and glaucomatous mice with OSK — no c-Myc. The mouse data is real. The human data does not exist yet.

Restoration — and the hype around it

This field now attracts billions of dollars, and with the money comes noise. Where the science ends and the marketing begins:

What "restoration" does NOT yet mean

  • No therapy has completed clinical proof of epigenetic age reversal in humans. Full stop.
  • A methylation clock moving in a small uncontrolled trial is a signal, not a result — clocks are proxies, and different clocks disagree.
  • Full reprogramming in the wrong dose causes teratomas — tumours of lost identity. The margin between "rejuvenated" and "no longer a liver cell" is the entire problem.
  • Supplements marketed as "epigenetic age reversal" have no controlled evidence behind the claim.

What the record DOES show

  • Methylation age is measurable, reproducible, and predicts mortality better than birthdays (Horvath 2013 and successors).
  • Partial reprogramming resets the clock in human cells (~30 years of reversal: Gill et al., Babraham Institute, eLife 2022) and restores function in mouse tissues.
  • Chemical reprogramming — small molecules instead of genes — exists (Hongkui Deng's lab, Peking University, Science 2013), hinting that a drug-like route is physically possible.
  • Serious capital agrees: Altos Labs launched in 2022 with ~$3 billion; Calico (Alphabet) since 2013; Retro Biosciences raised $180M from Sam Altman targeting +10 healthy years.

Human trials: the registry status

Everything above is cells and mice. What about humans? This page now carries the registry check itself — dated, scoped, and re-run quarterly.

One registered trial, zero completed — checked 2026-09-08

A search of ClinicalTrials.gov (API v2, 2026-09-08) for partial epigenetic reprogramming — by intervention, mechanism and sponsor terms — returns exactly one registered interventional human trial: NCT07290244, Life Biosciences' ER-100. An engineered AAV delivers the OSK trio (OCT4, SOX2, KLF4 — three of the four Yamanaka factors) into one eye of participants with open-angle glaucoma or NAION, with doxycycline switching the factors on for 56 days. It is a Phase 1, single-dose, open-label tolerability study: 18 participants estimated, recruiting since 2026-03-02, primary completion estimated May 2027. Registry record.

The same check finds no completed reprogramming trial, no posted efficacy or methylation-clock result, no Phase 2 or 3, and no systemic (whole-body) administration in humans. The first human experiment has begun; the first human result has not. Scope: this check covers ClinicalTrials.gov only — other national and WHO registries, company pipeline pages and preprints were not searched.

Panacea's position: modulation with designer molecules

The field's giant experiments use transcription-factor genes delivered by viruses — powerful, but hard to dose and hard to target. Panacea Bio Chem's research programme, led by Bogdan Dicoias, explores a complementary route: peptide- and enzyme-mediated epigenetic modulation — designer amino acid chain (AAC) molecules aimed at the writers, erasers and readers of the epigenome, where dosing can be titrated, stopped, and targeted tissue by tissue.

We state the boundary plainly: this is a research direction, not a product and not a therapy. Panacea claims no rejuvenation results. What we publish here is the scientific landscape and our position in it; programme details remain Panacea intellectual property until peer-reviewed disclosure.
